    綠茶調節脂肪細胞的功

    喝綠茶似乎有益於人體的健康,因為它與癌症、肥胖症、心臟血管疾病、血栓形成、高血壓、關節炎等生物醫學疾病之罹患率降低有關;也與體外培養癌細胞的死亡和生長抑制有關連。本實驗室最近的研究指出,綠茶唲茶素中epigallocatechin gallate(EGCG)可調節正常老鼠與肥胖老鼠的體重及其血脂含量,而結構相似的epicatechin gallate卻無法作用。本實驗室發現四種綠茶唲茶素中,EGCG會減少正在生長中和滿期中前脂肪細胞的數目,其效果較其它唲茶素強,同時顯示EGCG的作用濃度會因其劑量和細胞的發育狀態而異。EGCG減少前脂肪細胞的數目,可能起因於它抑制細胞的生長和造成細胞的自然致死,而此作用可能分別透過與細胞生長蛋白質激發酶(MAPK)和Caspase-3相關的途徑,其中以Erk MAPK為主,而非p38或JNK。EGCG也可透過抑制Cdk2蛋白質的表達量和活性、抑制cyclin D蛋白質的表達量、誘導Cdk2抑制蛋白質如p21和p27的表達量和調節這些蛋白質間的交互作用,進而造成前脂肪細胞的生長停滯。其次也發現EGCG會抑制胰島素、第一型和第二型類胰島素因子、或者雌性素促進前脂肪細胞的生長,而EGCG的抑制作用分別是改變各種荷爾蒙的訊息傳導途徑。例如EGCG會抑制胰島素接受體的活性、第一型類胰島素因子接受體的活性、第二型類胰島素因子接受體與其下游G蛋白質之間的交互作用、或與雌性素的非基因體途徑相關的MAPK的活性。最後則發現EGCG會調節脂肪細胞的分化指標,例如PPAR、C/EBPb、C/EBPa、三酸甘油等等。EGCG也會調節已分化的脂肪細胞的數目,只是所需的EGCG的劑量卻高於處理在前脂肪細胞的生長。不過,吾等發現EGCG處理在抑制前脂肪細胞的生長所需的劑量,低於處理在KB人類口腔癌細胞株和3T3纖維母細胞細胞株的生長。故本實驗結果,對於EGCG可調節動物的肥胖方面,可提供綠茶的作用機制與相關的科學線索。

    荷爾蒙與營養物調節抗胰島素激素的基因表達
    抗胰島素激素(resistin)是源於脂肪組織的新種多肽荷爾蒙,訊息核糖核酸和蛋白質的大小分別是約八百對核苷酸和一萬個道耳敦數。它一直被認為是肥胖症和糖尿病之間的橋樑,而此兩種疾病正是與高危險群的癌症、高血壓和心臟血管疾病息息相關。抗胰島素激素的命名,是因為它在影響動物對胰島素的忍受性和葡萄糖的忍受性,以及降低了胰島素刺激脂肪細胞攝入葡萄糖的能力。由於它在生物學上的重要性,最近逐漸開始研究其基因和蛋白質的表達之調控機制。雖然抗胰島素激素的基因和蛋白質表達可受到營養、內分泌、發炎、神經、遺傳、個體發育、藥物等相關因子調節,但是大部分的結果還未有定論,況且各因子的作用機制也不明瞭。故本實驗室分別探討類胰島素生長因子、雌性素和綠茶唲茶素、是否調節3T3-L1脂肪細胞內抗胰島素激素基因和蛋白質的表達,以及了解各類的作用機轉。本人選用這三大類的理由是因為根據先前的研究指出它們像抗胰島素激素和胰島素相類似,可調節3T3-L1脂肪細胞的活性和肥胖者的脂肪細胞活性。初步的結果發現,第一型和第二型類胰島素生長因子比胰島素較有效的抑制脂肪細胞內抗胰島素激素基因的表達,抑制的效果因作用時間和劑量而異。第一型類胰島素生長因子可加速抗胰島素激素mRNA的分解,其作用是透過第一型類胰島素生長因子接受器相關的途徑,但是與Erk, p38, PI3K等蛋白質激酶無關的途徑影響抗胰島素激素基因的表達。不過,第一型類胰島素生長因子可透過與p38蛋白質激酶相關的途徑影響抗胰島素激素抑制蛋白質的釋放。其次,我們發現綠茶唲茶素(俗稱維他命P)抑制抗胰島素激素基因的表達和減少細胞內蛋白質的含量,抑制的效果因處理的時間、劑量和唲茶素的種類而異。綠茶表沒食子唲茶素沒食子酸酯(EGCG)可增加抗胰島素激素mRNA的分解,它透過降低抗胰島素激素mRNA的穩定度而影響基因的表達,而此作用可能與Erk蛋白質激發酶相關途徑來調節抗胰島素激素mRNA的分解。最後,我們發現雌二醇可誘導脂肪細胞內抗胰島素激素基因的表達與蛋白質的釋放,刺激的效果因作用時間和劑量而異。它透過雌二醇接受器與C/EBPa蛋白結合影響抗胰島素激素的基因活性子(promoter)而降低抗胰島素激素的基因表達,其作用也與Erk相關但與p38和PI3K蛋白激酶無關的途徑。由於抗胰島素激素(resistin)可調節脂肪細胞的脂肪形成與增加細胞對胰島素的不靈敏,亦即及降低脂肪細胞攝入葡萄糖的能力,導致於糖尿病的產生。因此,本實驗室研究的成果關於類胰島素生長因子、雌性素和唲茶素可調節抗胰島素激素的基因表達與蛋白質釋放,首度架構三者和抗胰島素激素之間的橋樑和交互作用,並可對於綠茶唲茶素和類胰島素生長因子在抗胰島素激素誘導型的肥胖症和糖尿病的療程中是否成為有益的一種中草藥和荷爾蒙,可提供相關的作用機制與重要的科學數據參考。
